Background and Purpose— For years, the relationship between cardiac and neurological ischemic events has been limited to overlapping pathophysiological mechanisms and common risk factors. However, acute stroke may induce dramatic changes in cardiovascular function. Electrophysiological parameters of the myocardium are influenced by temperature. Working with experimental models of isolated heart, the analysis of modulating effect exerted by local changes of temperature can provide some information about the mechanisms of certain arrhythmias and their electrophysiological bases, such as ventricular fibrillation. In the two decades following Loewi’s work (8,9) on the frog heart several research groups tried to use the isolated heart preparation from higher vertebrates for further investigations on synthesis, release and inactivation of acetylcholine (ACh). Experiments in isolated cat hearts showed, that with increased Ca++-concentration of the perfusion-medium (0,56 g CaCl2/1000 ml) the onset of mechanical systole of left ventricle occurred earlier (21,9±3,3 msec after the onset ofQRS-complex in the ECG) than in experiments with a normal Ca++-concentration (0,28 g CaCl2/1000 ml). The onset of mechanical systole of the right ventricle was not effected.

Publisher Summary This chapter discusses the release of catecholamines from the heart. The isolated heart of the rabbit with the sympathetic nervous supply from the right stellate ganglion intact was set up according to Hukovio and Muscholl. The ganglion was stimulated for 30 s at supramaximal voltage with rectangular impulses of 3–5 ms duration a frequency of 10 shocks/s. Each stimulation period was followed by a resting period of 30 s. In the rabbit, four doses of 50 mg/kg l-α-methyldopa decreased the noradrenaline concentration in auricles, right, and left ventricles by 38–60%. Characterization of choline efflux from the perfused heart at rest and after muscarine receptor activation.

The resting efflux of choline from perfused chicken hearts varied from 0.4 to 2.6 nmol/g min, but was constant for at least 80 min in the individual experiments. The rate of choline efflux was found to be equal to the rate of choline formation in the heart, which, from the following reasons, was essentially due to hydrolysis of choline phospholipids. Cardiac content of choline phospholipids (7,200 nmol/g) was much higher than that of acetylcholine (5.5 nmol/g). Resting release of acetylcholine was 0.016 nmol/g min and, after inhibition of cholinesterase, only about 0.1 nmol/g min.
